Specificity:
VU1D9 reacts with the first EGF repeat in the extracellular domain of Ep-CAM. The epitope is sensitive for reducing agents. VU1D9 detects Ep-CAM in most simple. pseudostratified and transitional epithelia and in the majority of human carcinomas. VU1D9 is suitable for immunoblotting (non-reducing conditions). immunocytochemistry and immunohistochemistry on frozen and paraffin-embedded tissues and flow cytometry. Optimal antibody dilution should be determined by titration; recommended range is 1:25 ? 1:200 for flow cytometry. and for immunohistochemistry with avidin-biotinylated horseradish peroxidase complex (ABC) as detection reagent. and 1:100 ? 1:1000 for immunoblotting applications. Note:
Each vial contains 100 ?l 1 mg/ml purified antibody in PBS containing 0. 02% sodium azide. 0. 1% BSA and 50% glycerol. Ep-CAM is a 40 kD glycoprotein. which functions as an epithelial cell adhesion molecule. Ep-CAM mediates Ca2+-independent homotypic interactions. Formation of Ep-CAM-mediated adhesions has a negative regulatory effect on adhesions mediated by classic cadherins. which may have strong effects on the differentiation and growth of epithelial cells. Ep- CAM overexpression was suggested to be associated with enhanced epithelial proliferation. Ep-CAM is expressed in the majority of human carcinomas and is a marker for tumors of epithelial lineage.
