Note:
This product is an IgG fraction antibody purified from ascites or tissue culture supernate by affinity chromatography followed by extensive dialysis against the buffer stated above. Assay by SDS-PAGE resulted in a single protein band. The antibody reacts with DNP but not with carrier protein.
Application Note:
This monoclonal antibody reacts specifically with DNP (2 4-dinitrophenol). The antibody reacts with free DNP DNP conjugated to carriers such as thyroglobulin or BSA and detects the presence of DNP in proteins and nucleic acids. Although not tested this antibody is likely functional in RIA flow cytometry immunohistochemistry and immunoprecipitation. Recommended
Dilution:
This product was assayed against DNP conjugated BSA by ELISA using HRP Goatanti- Armenian Hamster IgG for detection. A working dilution of 1:2 000 to 1:10 000 is suggested for this assay. For immunoblotting dilute the antibody 1:1000 immediately before use. Researchers should determine optimal titers for other applications. Function:
Exhibits vasodilator natriuretic and diuretic properties in animal models and human tissues. Acts by stimulating cGMP via the natriuretic peptide receptor A (NPR1). Is a poor agonist of the atrial natriuretic peptide receptor B (NPR2). Is not degradated by neutral endopeptidase (NEP / MME). Binds to atrial natriuretic peptide clearance receptor (NPR3 / NPR-C) which may be responsible of the removal of DNP from the circulation. Increases calcium uptake and induces histamine release from rat peritoneal mast cells. Increases calcium-activated potassium current in gastric antral circular smooth muscle cells by increasing cGMP production and activating inositol trisphosphate receptors (IP3Rs).
