Immunogen:
Human Rhinovirus (HRV14) treated monocyte-derived dendritic cells.
Fusion Partner:
Spleen cells from immunised Balb/c mice were fused with cells of the X63-Ag8. 653 myeloma cell line.
Preparation:
Purified IgG prepared by affinity chromatography on Protein G from tissue culture supernatant
Buffer Solution:
Phosphate buffered saline pH7. 4

Flow Cytometry:
Use 10ul of the suggested working dilution to label 1x106 cells in 100ul. Suggested
Dilution:
Flow Cytometry - Neat - 1/10
Function:
Macrophage-restricted adhesion molecule that mediates sialic-acid dependent binding to lymphocytes including granulocytes monocytes natural killer cells B-cells and CD8 T-cells. Preferentially binds to alpha-2 3-linked sialic acid (By similarity). Binds to SPN/CD43 on T-cells (By similarity). May play a role in hemopoiesis.
Subcellular Location:
Isoform 1: Cell membrane; Single-pass type I membrane protein.
Subcellular Location:
Isoform 2: Secreted.
Tissue Specificity:
Expressed by macrophages in various tissues. High levels are found in spleen lymph node perivascular macrophages in brain and lower levels in bone marrow liver Kupffer cells and lamina propria of colon and lung. Also expressed by inflammatory macrophages in rheumatoid arthritis.
Similarity:
Belongs to the immunoglobulin superfamily. SIGLEC (sialic acid binding Ig-like lectin) family.
Similarity:
Contains 16 Ig-like C2-type (immunoglobulin-like) domains.
Similarity:
Contains 1 Ig-like V-type (immunoglobulin-like) domain. 1. Kirchberger. S. et al. (2005) Human rhinoviruses inhibit the accessory function of dendritic cells by inducing sialoadhesin and B7-H1 expression. 2. Hartnell. A. et al. (2001) Characterization of human sialoadhesin. a sialic acid binding receptor expressed by resident and inflammatory macrophage populations.
