IgG functions in the humoral immune response1. There are four subclasses of IgG, and they arenumbered according to decreasing abundance as IgG1 through IgG4. While IgG4 is the leastabundant of these subclasses, it is the predominant IgG involved in chronic or repeated antigenexposure, leading to an anti-inflammatory response. IgG4 is not usually part of the antibodyresponse to bacterial or viral infection. Rather, it dominates the response to allergens andparasitic worm infections. IgG4 has unique properties relative to other IgG antibodies1. IgG4 affinity for Fc receptors andcomplement is relatively lower and IgG4 is largely unable to activate antibody-dependent-immune effector responses. IgG4 also performs Fab-arm exchange and is more highlyglycosylated. Additionally, the IgG4 response is blocking in nature and able to induce toleranceand limit inflammation in the host, and particularly protects against allergic inflammation.IgG4 is produced by B cells that undergo class-switch recombination from IgM to IgG4 via Thelper 2 cell responses1.
