Description
DNA Marker 100bp DNA ladder is supplied at a total DNA concentration of 0.5mg/ml and is prepared from specially designed plasmid pME-80 DNA, containing pUC, lambda phage and yeast genome sequences.

The marker yields the following 10 discrete fragments (in base pairs): 1000, 900, 800, 700, 600, 500, 400, 300, 200, 100. Convenient for analysis of PCR products up to 1 kb and can be used for DNA quantification.

The bright band at 500 bp serves as the reference.

Concentration:
0.5mg DNA/ml

Storage Buffer:
10mM Tris-HCl, pH 7.6, 1mM EDTA.

6X Loading Dye Solution (L3550-01):
Supplied as a liquid in 10mM Tris-HCl, pH 7.6, 0.03% bromophenol blue, 0.03% xylene cyanol FF, 0.15% orange, 60% glycerol, 60mM EDTA.

Prepare DNA Marker for Agarose:
1-2ul DNA Ladder (0.5-1ug)
1ul of 6X Loading Dye Solution
3-4ul ddH2O

Prepare DNA Marker for Gels Polyacrylamide:
1-2ul DNA Ladder (0.5-1ug)
0.5ul of 6X Loading Dye Solution
0.5-1.5ul ddH2O

Protocol:
1. Mix gently
2. Load on the gel
*For gels with other lane widths, the comnponents of the mixture should be scaled either up or down. Use 0.2-0.4ul (0.1-0.2ug) of DNA Ladder oer 1mm of lane.

Recommendations:
1. Do not heat before loading.
2. Dilute DNA. sample with the 6X DNA. Loading Dye: mix 1 volume of the dye solution with 5 volumes of the DNA. sample. Load same volume of DNA sample and the DNA ladder.
3. Adjust concentration to equalize the amount of DNA. in the nearest band of the ladder.
4. For visualization with other intercalating dyes, do not add the dyes into the sample, use gel staining after electrophoresis or include dyes into agarose gel to avoid aberrant DNA migration.

Storage and Stability:
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 6 months after receipt at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Band Concentration:
0.5ug of DNA Ladder Mix contains:
a) 45ng (9.0%) for each of the 600-1000bp fragments.
b) 115ng (23.0%) for 500 bp band
c) 40ng (8.0%) for 100-400 bp bands.
