A reliable quantification of the extracellular matrix production is of central importance for example in fibrosis research, with collagen being one of the main matrix components. An increasingly popular assay is the hydroxyproline-based collagen assay. Hydroxyproline-based collagen tests measure hydroxyproline in hydrolyzed samples. One product highlight is the Total Collagen Assay Kit. It enables to determine collagen based on the quantitative colorimetric determination of hydroxyproline residues obtained by acid hydrolysis of collagen. In conventional commercial analyzes, HCl is removed after hydrolysis; this is a time-consuming step and requires special equipment. The QuickZyme Total Collagen Assay is the only hydroxyproline-based assay that bypasses this step. The result is a quick (<2 hours) and easy assay (96-well plate format) after the hydrolysis step. All types of collagen are recorded quantitatively, independently from species. Cell extracts, tissue homogenates or tissue per se are suitable as samples. The user friendliness here corresponds to that of ELISA.

Granzym B is a specific serine protease. It is found particularly in the intracellular granules of NK cells and cytotoxic T cells and plays a crucial role in the defense against viral infections. Granzyme B is encoded by the GZMB gene and is used in CD8-positive T cells for cytotoxicity in particular. It is stored in lytic granules together with perforins. Due to the prevailing conditions there, it is inactive. After a cytotoxic T cell gets into contact with a virus-infected cell, the granules are released and Granzyme B is activated. Activated granzyme B initiates apoptosis in the infected cell. The Granzyme B Activity Assay Reagent Set from QuickZyme Biosciences measures human active Granzyme B and also detects mouse Granzyme B. The reagent set for two 96-well plates provides quantitative evidence for numerous sample types, including body fluids (plasma, wound fluid), cell extracts and - culture media or tissue extracts. It is convenient to use like an ELISA and has the advantage of an overnight protocol. In this way a sensitivity of 0.01 ng / ml is achieved.
