Reaction Biology

Reaction Biology Corporation, headquartered in Malvern, Pennsylvania, was founded in 2001. The company specializes in providing comprehensive drug discovery services with a focus on biochemical and cellular assays, particularly in the fields of kinase profiling, epigenetics, and immune-oncology. Reaction Biology's expertise spans several therapeutic areas, assisting clients from the early discovery phase through to advanced drug development. The company has two main operational hubs for its global clients. Their primary headquarters and research centre are located in the US. Additionally, they operate a research site in Freiburg and Heidelberg, Germany, which complements their Malvern facilities.

The kinases that Reaction Biology provides are enzymes from the large family of protein kinases. These enzymes play a crucial role in the regulation of cellular processes by transferring phosphate groups to other proteins, a process known as phosphorylation. This is a central mechanism that modifies the function of proteins and influences various signalling pathways in the cell.

Serine/threonine kinases:

These kinases phosphorylate the amino acids serine or threonine in target proteins. They are important for processes such as cell growth and division. Well-known examples are the mitogen-activated protein kinases (MAPK) and protein kinase B (PKB/Akt), which are often investigated in cancer research as they are involved in cell proliferation and the survival of cancer cells.

Tyrosine kinases:

These kinases phosphorylate the amino acid tyrosine. Tyrosine kinases such as EGFR (Epidermal Growth Factor Receptor) and BCR-ABL are target structures in oncology as they are overactive in many tumour types. The activation of these kinases can lead to uncontrolled cell growth, which is why they are often blocked by specific inhibitors in order to stop tumour growth.

Lipid kinases:

A less common but still important group are kinases that phosphorylate lipids, such as PI3 kinase (phosphoinositide 3-kinase). These are important in signalling and influence cell processes such as growth and survival, which is why they also play a role in cancer research.
