Product Description
CRISPR-Cas12a (Cpf1) is a class II type V CRISPR system widely used for plant genome editing that relies on a short ~43 nt CRISPR RNA for DNA targeting 1. Cas12a proteins recognize T-rich protospacer adjacent motifs (PAM), with the canonical PAM being TTTV, and can be used in single gene or multiplex genome editing. Cas12a has RNase III activity 2 , enabling it to self-process the crRNA repeats, thereby enhancing its multiplexing ability 1. Cas12a cuts at distal sites from the PAM, generating staggered ends, and is more likely than Cas9 to cut and edit the target site repeatedly, which results in larger deletions or enhanced homology-directed repair than with Cas9 1. LbCas12a is derived from Lachnospiraceae bacterium ND2006 2.In plants, LbCas12a is the most commonly used CRISPR nuclease 1 and editing has been successfully performed in Arabidopsis, rice, maize, Nicotiana benthamiana, tomato, citrus, lettuce, cotton, poplar 1 , grapevine 2 , and soybean. Heat treatment can be used to improve the efficiency of LbCas12a editing 1, 2 . Applications include controlling gene expression via promoterdestruction 2.LbCas12a is also highly specific in humans and has an on-target efficiency similar to Streptococcus pyogenes Cas9 3. LbCas12a mediates robust genome editing for both single gene 4 and multiplex targeting 5 in HEK293FT cells and can efficiently induce both insertion and deletion mutations in U2OS human cells 3. The LbCas12a CRISPR system has also been used to develop a molecular diagnostic tool for point-of-care detection of SARS-CoV-2 nucleic acids 6. Additionally, LbCas12a variants are being explored as a potential therapeutic tool in the treatment of cancer and are able to induce efficient gene editing in oncogenes of human cells 7.
