IL-10 Protein, a major immune regulatory cytokine, exerts profound anti-inflammatory functions, limiting excessive tissue disruption caused by inflammation. Mechanistically, IL10 binds to its heterotetrameric receptor, IL10RA and IL10RB, leading to JAK1 and STAT2-mediated phosphorylation of STAT3, which translocates to the nucleus and drives the expression of anti-inflammatory mediators. It targets antigen-presenting cells like macrophages and monocytes, inhibiting their release of pro-inflammatory cytokines. Additionally, IL-10 interferes with antigen presentation, reducing MHC-class II expression, and controls macrophage inflammatory responses by reprogramming metabolic pathways.
