GSDMC protein, functioning as the precursor to the pore-forming protein, undergoes cleavage to release Gasdermin-C. This N-terminal moiety binds to membranes, forming pores with inner diameters of 10 to 15 nanometers and inducing pyroptosis. Initiated by caspase CASP8 cleavage of gasdermin-D in response to death signals, the cleaved protein moves to the plasma membrane, displaying robust binding to inner leaflet lipids, ultimately triggering pyroptosis.
