As a receptor for FN1, apolipoprotein APOE, and ALCAM/CD166, it inhibits receptor-mediated phosphorylation, intracellular calcium mobilization, and FCGR1A/CD64-mediated monocyte activation, reducing TNF production. It impedes T cell proliferation, induces anergy, suppresses IFNG-producing CD8+ cytotoxic T cells, promotes CD8+ T suppressor cells, up-regulates CD86 on dendritic cells, interferes with TNFRSF5-signaling, and inhibits NF-kappa-B up-regulation. Interactions with FN1 and phosphatase PTPN6 mediate these inhibitory effects.
