Purification	HeLa cells, tansfected with human HE4 overexpression vector and empty vector respectively, are harvested and washed with PBS. The cells were further lyzed with Raybiotech Mammalian Cell Lysis Buffer (compatible with most enzyme assays) containing mammalian cell protease inhibitor cocktail. After clarified with 20, 000 g centrifugation for 30 min, the lysate protein concentration was determined by BCA kit (Thermo Scientific Inc.). The lysates were aliquoted and stored at , 80 , C immediately.

Reconstitution	Briefly spin the vial and bring the contents to the bottom prior to opening. It is recommended to reconstitute at 0.5~1 mg/mL with sterile deionized water.
