Description: Rottlerin (NSC-56346; NSC-94525), a naturally occuring compound isolated from Mallotus Philippinensis with the potential to be used as a chemotherapeutic agent for adrenocortical carcinoma, is a novel and specific protein kinase C (PKC) inhibitor with IC50 values for PKCdelta of 3-6 uM, PKCalpha, beta, gamma of 30-42 uM, PKCepsilon, eta, zeta of 80-100 uM. Rottlerin acts as a direct mitochondrial uncoupler, and stimulates autophagy by targeting a signaling cascade upstream of mTORC1. Rottlerin induces apoptosis via caspase 3 activation. Rottlerin is an angiogenesis inhibitor and an inhibitor of protein kinase Cdelta (PKCdelta) and calmodulin kinase III.
