Origin:

Animals were immunized with a transfected NIH 3T3 cell line, 18-3-7, that expresses full length normal human neu protein. Spleen cells were fused with SP2/O myeloma cells. The antibody recognizes substantially purified p100, human neu related protein, the full-length HER-2/neu molecule in tumor tissue (p185) or the extracellular domain (ECD) (p105) in serum, plasma, cell cultures and fluids.

AddexBio-formulated DMEM (C0003-01) , or equivalent medium + 10% FBS

Subculture:

Cultures can be maintained by addition or replacement of fresh medium. Start cultures at 2 X 10^4 cells/mL and maintain between 3 X 10^5 and 5 X 10^5 cells/mL. Do not allow the cell concentration to reach 1 x 10^6 cells/mL. Population doubling time: 34 hours. Culture at 5% CO2, 37C.

Freezing Medium:

Complete culture medium supplemented with 7.5% (v/v) DMSO
