ADAM10, which is known as a disintegrin and metalloprotease 10, is a cell surface protein enconded by the ADAM10 gene. ADAM10 is a sheddase which cleaves membrane bound proteins at cell surface. After cleavage, the sheddase releases ectodomains with modified location and function. ADAM10 cleaves ephrin, and when this happens, the entire ephrin/eph complex is endocytosed. ADAM10 also has aplha secretase activity which allows processing of amyloid precursor protein. Inhibition of ADAM10 in breast cancer patients shows novel results as ADAM10 shows association with HER2 shedding. The inhibition of ADAM10 decreases HER2 cell lines proliferation.
