Biological Activity	COH34 is a potent and specific poly(ADP-ribose) glycohydrolase (PARG) inhibitor with an IC₅₀ of 0.37 nM. COH34 binds to the catalytic domain of PARG (K_d=0.547 uM), thereby prolonging PARylation at DNA lesions and trapping DNA repair factors^[1]. IC50 & Target: IC50: 0.37 nM (PARG)^[1] In Vitro: COH34 efficiently kills PARP inhibitor-resistant cancer cells^[1]. In Vivo: COH34 induces lethality in cancer cells with DNA repair defects and exhibits antitumor activity in xenograft mouse cancer models^[1].
