Reconstitution
Centrifuge the vial prior to opening. Reconstitute in sterile PBS, pH 7.4 to a concentration of 50 µ g/ml. Do not vortex. This solution can be stored at 2-8° C for up to 1 month. For extended storage, it is recommended to store at -20° C.

Biological activity
Measured by its ability to inhibit trypsin cleavage of a fluorogenic peptide substrate, Mca - RPKPVE - Nval - WRK (Dnp) - NH2. The IC50 value is < 2.5 nM, as measured in 100uL reaction mixture containing 1.25 ng trypsin, 10 uM substrate, 50 mM Tris, 10 mM CaCl2, 0.15 M NaCl, 0.05% Brij-35, pH 7.5.
Introduction
Tissue Factor Pathway Inhibitor 2 (TFPI­2), also known as placental protein 5 (PP5) and retinal pigment epithelial cell factor 1 (REF­1), is a member of the Kunitz-type serine proteinase inhibitor family, is a structural homologue of tissue factor pathway inhibitor (TFPI). TFPI2 is highly abundant in the full-term placenta and widely expressed in umbilical vein endothelial cells, liver, placenta, heart, pancreas, and maternal serum at advanced pregnancy. TFPI­2 may play a role in the regulation of plasmin-mediated matrix remodeling. Inhibits trypsin, plasmin, factor VIIa/tissue factor and weakly factor Xa and has no effect on thrombin. Reduced synthesis of TFPI-2 has been related to numerous pathophysiological processes such as inflammation, angiogenesis, atherosclerosis, retinal degeneration and tumor growth / metastasis.
